Cervical cancer can be effectively prevented through screening and HPV vaccination. In the context of many countries adopting a single-dose schedule, expanding the target age range, and adjusting immunization regimens, the development and standardization of immunogenicity assessment methods are essential for comparing different vaccines and vaccination schedules, as well as for supporting post-implementation surveillance and updating recommendations. We conducted this study with the aim of initially establishing and applying methods for evaluating anti-HPV antibody immunity. In this study, we established enzyme-linked immunosorbent assay (ELISA) and Luminex immunoassay methods to detect and quantify antibodies against HPV16, and compared their performance with the gold standard HPV neutralization assay developed by the Murdoch Children’s Research Institute, using serum samples collected before and after vaccination. The results demonstrated strong correlations, with R2 values of 0.92 and 0.96 for the neutralization–Luminex and neutralization ELISA comparisons, respectively. These findings suggest that ELISA and Luminex assays are reliable alternatives to neutralization assays for assessing immune responses to HPV16, with promising potential for broader application to other HPV types.
Biography of the presenting author
Le Thi Khanh Ly is the Head of the Cell Technology Laboratory at the Center for Biomedical Research, National Institute of Hygiene and Epidemiology, Hanoi, Vietnam. She obtained her Bachelor’s and Master’s degrees in Biology from the VNU University of Science, Vietnam National University, Hanoi, in 2019 and 2022, respectively. Her research focuses on virology, immunology, and the molecular epidemiology of infectious diseases, with particular interests in rotavirus, calicivirus, and human papillomavirus (HPV). She has extensive experience in molecular and immunological techniques, including PCR-based assays, ELISA, flow cytometry, and next-generation sequencing.
She has participated in multiple international research projects and clinical trials related to vaccine effectiveness and infectious disease surveillance, in collaboration with global partners such as the US CDC and the National Institute of Infectious Diseases (NIID), Japan, among others. She has authored and co-authored more than 26 publications and has presented her work at several international conferences.
